JACK & JILL was a romantic drama about life and loves of young people in New York, Jacqueline "Jack" Barrett and David "Jill" Jillefsky, and their friends, Mikey Russo, Barto Zane, Elisa Cronkite and Audrey Hriffin. The show follows their lifes after college, as they are trying to reach over to the adult life, forgetting about their childhood, exploring their sexuality, and finding the difference between a fairy tale and real life.

Meanwhile, Barto's grades suffer because he can't stop thinking about his one-night stand with Audrey; Audrey begins to doubt her relationship with Travis; and Mikey attends a singles sminar with Elisa.

Jill's younger sister Abby comes to visit. Elisa goes to dinner and to a play solo. Audrey and Barto sleep together. Mikey takes Abby to a night club and Jill goes to find her. Jill and Abby have a fight but later part on good terms. Barto sees Audrey hanging out with Travis, the director again and he is crushed.

Jill's boss, Mr. Ashton dies before he can give Jill a promised promotion. Jill insults his new boss during a business meeting. Jack takes a job at a dirty restaurant in order to get a story on poor restaurant sanitation. Audrey is afraid to break up with her director boyfriend after he gets her an important role. Jack tries to help Elisa and Jill get back together but Elisa can't help feeling that there is something between Jack and Jill. Jill contemplates leaving Major Toys but later makes peace with the new boss, Theodore.

With just three weeks to go before Jack and Jill's wedding, issues start to pop up that reveal how little the couple has discussed life's big issues with each other. So, after a discussion with Elisa, Jack draws up a list of topics to go over, and the result is a fight with Jill, and Jack going off to scout the Bahamas wedding location alone, for ""some needed time apart"". Barto is forced to admit to Emily that the reason he and Audrey broke up is because Barto was unfaithful. Audrey moves back to her place (with Russell & Kate having departed), and lets her agent talk her into going on a 'Celebrity Date' game show. Audrey's date seems harmless enough at first—until she finds him inside her apartment, uninvited! A restraining order later, Audrey is forced to move back in with Elisa-Mikey-Barto.

After a customer causes a disturbance at the bar, Dante hires a bouncer without consulting Jill or Mikey. Jack sets out to mend her strained relationship with Elisa. Elsewhere, Audrey has an embarrassing encounter with Russell, and Barto flirts with a pretty waitress at a café.

Jack and Jill scramble to keep their wedding on track after the hurricane destroys their Bahamas plan, but the couple is in a bad place, something that is especially concerning Jill. Jack talks things over with Jill's mom, and Jill talks things over with Elisa. Eventually, Jill asks Jack to forget the long-term wedding arrangements, and to just get married the next day at the Justice of the Peace. Meanwhile, Mikey and Elisa plan a party for their 'unattached' single friends, but they are shocked to learn just how depleted that group of friends has become, as most of their friends are now paired up! And Jack & Jill's sudden wedding plans force them to change the party into a wedding party anyway. An overheard comment from Barto about how Audrey should face her fears vis-a-vis her stalker incident prompts Audrey to move back to her place. The next day, at Major Toys, Jill's boss announces that he is putting one of Jill's creations, Zantopia, into development.
