Noah's Arc follows the lives of four African-American men in Los Angeles. Noah, the protagonist of the series, is a struggling screenwriter who falls for Wade, a fellow screenwriter who is questioning his sexuality. Outspoken HIV counselor Alex has some self-confidence issues and is struggling to hold onto his sexy anesthesiologist boyfriend, Trey. Ricky is the promiscuous owner of a clothing store on Melrose Avenue. Economics professor Chance struggles to maintain his sanity after moving in with his boyfriend Eddie and Eddie's daughter, Kenya.

Alex shocks his friends by deciding to sign up for an amateur drag competition. But he gets cold feet before going up on stage. Noah wants to meet Wade's straight friends but he feels uncomfortable by Wade's attempts to change his appearance. Ricky tries to prove to his friends that he isn't addicted to sex. Chance tries to get over Eddie but is consoled by a stranger.

Meanwhile, Noah faces a dilemma when Brandy, the Paramount executive whom Noah once had an aborted threesome with Wade, wants him to edit a script by Wade. Meanwhile, Alex comes to a major decision about his career when he leaves his job at the HIV/AIDS clinic.
